  • 生产达菲的关键原料:莽草酸是罗氏生产达菲的关键原料。目前罗氏使用的莽草酸是从八角中加工提取的。 从石油化工原料合成莽草酸步骤复杂,目前除科研工作外没有生产和应用的价值。莽草酸可以从另外一种天然产物奎宁酸合成,需要几步反应。奎宁酸来源于金鸡纳树(cinchona tree)的树皮,可以像莽草酸一样提取和大量供应。金鸡纳树的产地主要在非洲国家扎伊尔(Zaire),但由于扎伊尔的政局不稳定,难以在商业上确保金鸡纳树树皮的供应。 目前另外一个生产莽草酸的方法是采用生物发酵的方法制备,以葡萄糖为原料。这是罗氏在达菲的生产中为防止八角(莽草酸)来源不稳定而准备的第二套生产方案。 罗氏的发酵法莽草酸是在北欧生产的。采用发酵法生产莽草酸的厂家曾由于生产成本太高造成亏损而停止生产。罗氏自称,其用于达菲生产的莽草酸有三分之一来源于发酵法生产。
